Stock Investing Basics Dichlorophenol-indophenol (DCIP) precipitation test is a very simple and useful test for detection of unstable hemoglobin such as hemoglobin E (Hb E) and hemoglobin H (Hb H). DCIP is a synthetic dye which has deep blue color in water. Interpretation of precipitation or cloudy appearance in such dark blue solution is very frustrating especially for those who do not have experiences. The present study is the first report to modify the test by adding 40 M ascorbic acid to clear the deep blue color. Quantitation of the hemoglobin could be done by measuring the absorbance at 800 nm. It was found that the absorbance of homozygous Hb E, heterozygous Hb E and Hb A2A were 1.01 + 0.35, 0.39 + 0.09 and 0.1 + 0.05, respectively . The absorbance was consistent with the level of Hb E determined by hemoglobin electrophoresis and high performance liquid chromatography (HPLC). Share this:
